Abstract: Seit dem Nachweis von Clofibrinsäure in vielen aquatischen Systemen um Berlin Anfang der 90er Jahre ist die Anzahl der nachgewiesenen Arzneimittelrückstände kontinuierlich gestiegen. Dieser Arzneimitteleintrag in die Umwelt führte sowohl in aquatischen als auch in nichtaquatischen Systemen zu schweren Schädigungen der Fauna. Im Rahmen dieser Dissertation wurde eine Methode zur Bestimmung von 16 wichtigen Arzneimittelwirkstoffen aus den Gruppen der Analgetika, der Lipidsenker, der Hormone, der Psychopharmaka und der Phosphodiesterase-Typ5-Hemmer mittels LC-MS/MS entwickelt. Zur Kompensation von Matrixeffekten und Verlusten bei der Aufarbeitung wurde für die Quantifizierung bei der negativen Ionisation 37Cl-markiertes Chloramphenicol und bei der positiven Ionisation deuteriertes Diazepam verwendet. So konnte für 13 von 16 Analyten eine Bestimmungsgrenze unterhalb des Schwellenwertes für die ökotoxikologische Bewertung von 10 ng/L erreicht werden. Mit der entwickelten Methode wurden Proben aus der Saale und Seen um Halle vermessen. Hierbei konnte bestätigt werden, dass Carbamazepin (Medianwert 202,5 ng/L) und Diclofenac (Medianwert 166,9 ng/L) an vielen Stellen der Saale nachgewiesen wurden und der Abwasserpfad die wichtigste Eintragsquelle für Arzneimittelwirkstoffe in die Umwelt darstellt.
The number of pharmaceutical residues found in aquatic systems has been increasing since the detection of Clofibric acid around Berlin at the beginning of the 1990s. This enter into the environment led to heavy fauna damage in aquatic as well as in nonaquatic systems. An LC-MS/MS method to determine 16 important pharmaceuticals belonging to analgetics, antilipidemic drugs, hormones, psychotropics, phosphodiesterase- typ5-inhibitors was developed in this thesis. 37Cl-marked Chloramphenicole and deuterated Diazepam were used for quantification in negative and positive mode respectively, to compensate for matrix effects and loss during sample preparation. So a limit of quantification below the threshold for ecotoxicological assessment of 10 ng/L was achieved for 13 out of 16 analytes. Samples from the river Saale and lakes around Halle were quantified with the method. It was confirmed that, Carbamacepin (median 202.5 ng/L) and Diclofenac (median 166.9 ng/L) are found at various points in the aquatic environment of the Saale and that the wastewater path is the most important source for contamination with pharmaceutical residues.
