Abstract: Das computergestützte Wirkstoffdesign ist zu einem festen Bestandteil der Arzneimittelforschung des 21. Jahrhunderts geworden, deren Anwendung sich von der Identifizierung erster potentieller Ausgangsverbindungen bis hin zur Optimierung schon bestehender Leitstrukturverbindungen erstreckt. In dieser vorliegenden Arbeit wird gezeigt, wie potente und selektive Inhibitoren der Histondeacetylase (HDAC) 8, 6 und 11 durch das Anwenden verschiedener theoretischer computergestützter Methoden erfolgreich analysiert und entwickelt werden konnten. Diese epigenetischen HDAC-Enzyme gelten als vielversprechende Zielstrukturen bei der Behandlung unterschiedlicher Erkrankungen, in deren insbesondere Subtyp-selektiven Hemmung großes pharmazeutisches Potential gesehen wird.
Computer-aided drug design has become an integral part of pharmaceutical research in the 21st century, with applications ranging from the identification of first potential starting compounds to the optimization of existing lead structures. In this thesis, it is shown how potent and selective inhibitors of the histone deacetylase (HDAC) 8, 6 and 11 could be successfully analyzed and developed by using different theoretical computer-aided methods. These epigenetic HDAC enzymes are considered to be promising target structures in the treatment of various diseases, and their subtype-selective inhibition, in particular, is expected to have great pharmaceutical potential.
