Abstract: Im Vergleich zu Hefe und tierischen Modellsystemen steht die Untersuchung von Kaskaden der MAP-Kinasen und den dazugehörenden Regulationsmechanismen in Pflanzen noch am Anfang. Da auch Protein-Protein-Interaktionen als molekulare Basis der Regulation von MAPK-Kaskaden dienen, war es das Ziel der vorliegenden Arbeit, MAPK-Proteinkomplexe aus Arabidopsis thaliana mittels Tandemaffinitätsreinigung zu isolieren. Zuerst wurde die Möglichkeit für einen immunologischen Nachweis der MAPK3 und 6 aus Arabidopsis thaliana geschaffen. Zudem konnten mittels Ausschlusschromatographie Hinweise über die Anwesenheit von AtMPK3 und 6 in Proteinkomplexen erbracht werden. Die Optimierung der Aufreinigung von AtMPK3-TAP gestaltete sich schwierig. So war die Anreicherung von markierter AtMPK3 deutlich geringer als die Anreicherung des TAPmarkierten Transkriptionsfaktors AtTGA2. Mit Hilfe verschiedener Experimente konnte nicht geklärt werden, ob der große Verlust an AtMPK3-Protein auf eine Adsorption an die verwendeten Materialien und/oder auf eine Proteolyse zurückzuführen war. Erste Versuche, durch eine einstufige Affinitätsreinigung die Effizienz der Anreicherung von AtMPK3 zu verbessern und damit die Chancen für eine Identifizierung von eventuell mit AtMPK3 assoziierten Proteinen zu erhöhen, schlugen fehl. Obwohl die Eluate nach der zweistufigen Affinitätschromatographie noch komplexe Proteinmischungen darstellten, waren nach der Auftrennung im denaturierenden Gradientengel und der Silberfärbung der Proteine Unterschiede in der Zusammensetzung der Proteinproben nachweisbar. In einem Fall ließen sich im Anschluss an die MALDI-TOF MS-Analyse und die Datenbanksuche mit MASCOT mehrere der identifizierten Peptide der Sequenz von AtMPK3 zuordnen. Somit gelang zwar der massenspektrometrische Nachweis eines niedrig abundanten Proteins wie AtMPK3 in einer aus Blattmaterial erhaltenen Probe, jedoch nicht die Isolierung und Identifizierung von putativ mit AtMPK3 interagierenden Proteinen. Es bleibt zu klären, ob mit anderen Techniken der Proteomik nach einer affinitätschromatographischen Reinigung von AtMPK3 putative Interaktoren nachweisbar sind.
The study of MAPK cascades and their regulation in plants is just at the beginning compared to what is know about the MAPK networks in yeast and animals. Since protein-protein interactions play an important role as molecular base for the regulation of MAPK cascades, the aim of this work was to isolate MAPK protein complexes from Arabidopsis thaliana by tandem affinity purification (TAP). In the first place antibodies were prepared to be able to detect MAPK3 and 6 from Arabidopsis thaliana by Western Blotting. Moreover, by using size exclusion chromatography it was possible to obtain indications of the presence of AtMPK3 and 6 in protein complexes. The optimisation of the purification of AtMPK3-TAP turned out to be difficult. The enrichment of AtMPK3 protein was significantly lower than the enrichment of the TAP-labelled transcription factor AtTGA2. By means of different experiments it was not possible to clarify, if the loss of AtMPK3 protein was due to the adsorption to the materials used and/or due to proteolysis. So far, the reduction of the loss of AtMPK3 protein by using one step instead of two step affinity purification failed. In addition, the eluats resulting from tandem affinity purification of AtMPK3 were complex mixtures of proteins. Nevertheless it was possible to identify differences in the composition of the eluates of the control compared to the purification of AtMPK3 after separation on a gradient gel and staining with silver. In one case peptides of AtMPK3 could be identified by MALDI-TOF MS analysis and data bank search using MASCOT. Thus, the isolation and identification of a low abundance protein like AtMPK3 from leave material after affinity purification combined with mass spectroscopy was successful. However, no putative interactors of AtMPK3 could be identified. It remains to be clarified if other techniques used in proteomics allow the identification vof putative interactors of AtMPK3.
