Abstract: Die Arbeit sollte einen Beitrag zur weiteren Erforschung der Cytochrom P450-abhängigen Enzymaktivitäten in HaCaT-Keratinozyten leisten. In einem ersten Teil wurden alle relevanten Substanzen, die mit den Zellen in Berührung kamen, auf ihre Zytotoxizität untersucht. Es wurden dabei Studien zur membranschädigenden Wirkung mit dem Neutralrot-Test (NR-Test) durchgeführt und mitochondriale Schädigungen mit dem MTT-Test erfasst. Die Bestimmung der Enzymaktivität von Phase-I-Reaktionen erfolgte mit so genannten Enzymvermittelten Indikatorreaktionen, die die Aktivität einzelner Enzyme repräsentieren. Der Schwerpunkt der vorliegenden Arbeit lag in der Charakterisierung der Enzyme CYP1A1/2 und 1B1, der für den Arzneistoff-Metabolismus relevanten Enzyms CYP 3A3/4/5 und der Subfamilie 4A. Folgende Reaktionen wurden untersucht: 7-Ethoxyresorufin-O-Deethylierung (EROD); 7-Ethoxycoumarin-O-Deethylierung (ECOD); Testosteron-Metabolisierung und 4-(N,N-Dimethylamino)azobenzen-Reduktion (DAB). Es wurde die Beeinflussung der Metabolisierung der Substrate 7-Ethoxyresorufin, 7-Ethoxycoumarin, Testosteron und 4-(N,N-Dimethylamino)azobenzen unter dem Einfluss von Induktoren und Inhibitoren untersucht. Außerdem wurde untersucht, ob eine UVA- oder UVB-Bestrahlung der HaCaT-Zellkultur vor der Bestimmung der enzymatischen Aktivität Einfluss auf die metabolischen Prozesse hat.
The aim was to make a contribution to the research of cytochrom P450-dependent enzyme activity in HaCaT-keratinocytes. In the first part all relevant substances, witch were in contact with the cells has been investigated because of there cytotoxicity. Studies were made to demonstrate the toxic effect against membranes with the neutralred-assay (NR-Assay) and second the mitochondrial-toxicity was investigated with the MTT-Assay. The investigation of phase I enzyme activities of was made with so called enzyme-mediated indicator-reactions, witch are representing the activity of individual enzymes. The main emphasis of this investigation was to characterize the enzymes CYP1A1/2 and 1B1, the enzymes CYP 3A3/4/5, witch are relevant for drug metabolisation, and last subfamily 4A. In this connexion were investigated the 7-ethoxyresorufin O-deethylase (EROD); the 7-ethoxycoumarin O-deethylase (ECOD), the testosterone metabolism and the Azoreduction of dimethylaminoazobenzene (DAB). The influence of certain inductors and inhibitors on metabolism of 7-ethoxyresorufin, 7-ethoxycoumarin, testosterone and dimethylaminoazobenzene was examined. Furthermore was investigated, if were is an influence of UVA- and UVB-irradiation on the enzymatic activity in HaCaT-cells before the enzyme-mediated indicator-reactions started.
