Abstract: Während der embryonalen Spezifizierung hypaxialer Skelettmuskulatur kommt es zur Delamination, Migration und zur myogenen Differenzierung von Muskelvorläuferzellen. Nach Überexpression in ex vivo Gewebe-Kulturen waren Lbx1 und Pax3 in der Lage, die Transkription von MyoD, Myogenin und MHC zu induzieren. Zusätzlich wurde eine wechselseitige Regulation von Pax3 und Lbx1 festgestellt. Pax3 konnte als notwendiger Faktor zur transkriptionellen Aktivierung von Lbx1 identifiziert werden. Nach erfolgter Lbx1 Aktivierung konnte Pax3 als hinreichender - aber nicht weiter notwendiger Faktor zur Aufrechterhaltung der Lbx1 Transkription ermittelt werden. In Somiten-Kulturen steigern Lbx1 und Pax3, retroviral überexprimiert, die Proliferationsrate um ein vielfaches. Die Blockierung des Zellzyklusses bei gleichzeitiger retroviraler Lbx1 oder Pax3 Infektion führt zu einem Verlust der Induktion der myogenen Differenzierung. Im zweiten Teil der Arbeit wurde eine heterozygot vorliegende Mutation des Transkriptionsfaktors und myogenen Differenzierungsgens Myf-6 des Menschen analysiert. Die biochemische Mutationsanalyse und die im Mausmodell nachgestellten Genotypen deuten auf eine haploide Insuffizienz des Myf-6 Gens hin, die zur Ausbildung einer milden Myopathie führt. Dagegen zeigt die Kombination aus heterozygoter Myf-6 Mutation und einer Dystrophinopathie des Typs Becker (BMD) einen Synergismus beider Mutationen auf, durch den die resultierenden massiven Regenerationsdefekte einen besonders schweren Krankheitsverlauf aufweisen.
In the present study we have investigated the roles of Lbx1 and Pax3 in specifications of hypaxial muscle development. It was shown that infection of embryonic tissues with a retrovirus encoding Lbx1 or Pax3 is sufficient to induce muscle differentiation monitored by expression of MyoD, Myogenin and myosin heavy chain. Lbx1 and Pax3 were able to regulate their transcription reciprocally. Pax3 and Lbx1 were identified as keyregulators of hypaxial myogenesis. Furthermore Lbx1 and Pax3 function as stimulating factors to amplify the myogenic precursor cell pool with their increasing potential of myogenic differentiation. In the second project we investigated a human detected heterozygous mutation in the myogenic factor Myf-6. Myogenic factors (MYF) belong to the basic helix-loop-helix (bHLH) transcription factor family and regulate myogenesis and muscle regeneration. Protein-protein interaction of mutant MYF6 was reduced, and DNA-binding potential and transactivation capacity were abolished.The mutated Myf-6 protein is most likely due to haploinsufficiency and causes a mild myopathy. The combination of this Myf-6 mutation with a Becker muscular distrophy aggravates the clinical course of this dystrophinopathy.
