Abstract: Der Comet Assay ist eine mikroelektrophoretische Methode zur Bestimmung genotoxischer Effekte. Der Anteil im elektrischen Feld mobiler Kern-DNA, die den Comet-Schweif bildet, wird fluorimetrisch ermittelt und stellt ein Maß für Strangbrüche in der untersuchten DNA dar. In dieser Arbeit sind Versuchsbedingungen ermittelt worden, die den Einsatz des Comet Assay für die Genotoxizitätstestung an Pflanzen (Vicia faba, Hordeum vulgare, Arabidopsis thaliana) effizient und reproduzierbar ermöglichen. Drei Protokollvarianten des Comet Assay (A/A, A/N, N/N) sind zu diesem Zweck für pflanzliche Zellkerne etabliert worden. DNA-Effekte von Genotoxinen verschiedener Wirkgruppen (MMS/MNU, MH, Bleomycin, Mitomycin C) sowie Reparaturverläufe nach Alkylantien- bzw. Bleomycin-Behandlung wurden vergleichend untersucht. Mit dem Comet Assay konnten auch adaptative Vorgänge, vergleichbar der "Clastogenen Adaptation", erfasst werden. Der Comet Assay wurde zudem mit Fluoreszenz in situ Hybridisierung (FISH) sowie Durchflusscytometrie kombiniert. Dadurch wurde die Lösung weitergehender Fragestellungen hinsichtlich der Beteiligung spezifischer Chromatin-Domänen an der Schweifbildung, des Einflusses der Zellzyklusphase auf die DNA-Migration sowie der Enstehung von Aberrationsclustern in Heterochromatinbereichen ermöglicht.
The comet assay is a micro-electrophoretic method for the evaluation of genotoxic effects. The portion of nuclear DNA which migrates towards the anode forming the comet tail is captured fluorometrically as a measure for DNA damage. This thesis describes experiments to establish applicable conditions for efficient and reproducible genotoxic testing with the comet assay in plants using Vicia faba, Hordeum vulgare and Arabidopsis thaliana. To this end three variants of the comet assay protocol (A/A, A/N and N/N) were introduced for plant cell nuclei. DNA effects of chemicals belonging to different classes of mutagens (MMS/MNU, MH, Bleomycin, Mitomycin C) as well as repair kinetics after alkylating and bleomycin treatment, respectively, have been investigated. The comet assay revealed also adaptive effects comparable to "clastogenic adaptation". Additionally, the comet assay was used in combination with fluorescence in situ hybridization (FISH) and flow cytometry. Thereby questions regarding the involvement of specific chromatin domains in tail formation, the influence of cell cycle stage on DNA migration as well as the origin of aberration clusters in heterochromatic regions could be solved.
