Abstract: Die 3'-Enden eukaryotischer mRNAs entstehen durch posttranskriptionelle Prozessierung von prä-mRNAs. Während der Polyadenylierung der prä-mRNAs in Säugerzellen wird die Poly(A)-Polymerase durch das nukleäre Poly(A)-Bindungsprotein 1 (PABPN1) stimuliert. Dies führt zu einer etwa 30-fach höheren Affinität der Poly(A)-Polymerase für die RNA, ohne dass PABPN1 direkt in die Katalyse eingreift. Für die Stimulierung der Poly(A)-Polymerase werden nicht nur die zwei RNA-Bindungsdomänen von PABPN1 benötigt sondern auch eine α-helikale Region ähnlich einer coiled-coil-Domäne im N-Terminus des Proteins. Es wurden eine Reihe Punktmutanten in der α-helikalen Region hergestellt. Die Mutanten wurden hinsichtlich RNA-Bindung und Stimulierung der Poly(A)-Polymerase getestet. Die Punktmutante L136S war nicht in der Lage, die Poly(A)-Polymerase zu stimulieren. Für die Längenkontrolle der Poly(A)-Schwänze von prä-mRNAs sind drei Proteine nötig: die Poly(A)-Polymerase, der cleavage and polyadenylation specificity factor (CPSF) und PABPN1. Auf vollständig verlängerten Poly(A)-Schwänzen, welche vollständig mit PABPN1 bedeckt sind, wurden Wechselwirkungen zwischen der Poly(A)-Polymerase und CPSF unterbrochen. Aus diesen Daten wurde ein Modell für die Längenkontrolle der Poly(A)-Schwänze aufgestellt.
3'ends of eukaryotic mRNAs are generated by post-transcriptional processing of precursor RNAs. During the polyadenylation of mRNA precursors in metazoan cells, poly(A) polymerase is stimulated by the nuclear poly(A) binding protein, PABPN1. Binding of PABPN1 to the substrate RNA is required for the stimulation of poly(A) polymerase and leads to a around thirty-fold increase in the apparent affinity of poly(A) polymerase for the RNA without significant effects on the catalytic efficiency. The stimulation requires not only the two RNA binding domains of PABPN1 but also an α-helical region resembling a coiled-coil domain. A set of point mutations in this α-helix were made and tested for RNA binding and stimulation of poly(A) polymerase. The point mutation L136S was sufficient to block the stimulatory activity. Three proteins are required for length control of poly(A) tails: poly(A) polymerase, cleavage and polyadenylation specificity factor (CPSF) and PABPN1. Interactions between CPSF and poly(A) polymerase disappear on full elongated poly(A) tails covered with PABPN1. A possible mechanism for length control was postulated.
