Abstract: Die Spaltung des Donorsubstrates D-Xylulose-5-phosphates durch Wildtyp-Transketolase und die Transketolasevariante H263A (TK-H263A) wurde mit Hilfe von Enzymkinetik und CD-Spektroskopie untersucht. Beide Enzyme spalten das Donorsubstrat auch in Abwesenheit eines Akzeptorsubstrates. Dabei ist die Spaltung des "aktiven Glycolaldehyds" (α-Carbanion/Enamin Intermediat) bei der Variante H263A beschleunigt. Chemisch synthetisiertes Intermediat D, L-(α,β-Dihydroxyethyl) Thiamindiphosphat (Racemat) bindet an Wildtyp-Transketolase mit einem vergleichbaren KD wie der native Kofaktor. Beide Enantiomere werden vom Enzym, allerdings mit unterschiedlichen Geschwindigkeiten, gespalten. Im Gegensatz zum enzymatisch generierten α-Carbanion von a,b-Dihydroxyethyl-Thiamindiphosphat, welches nach Decarboxylierung von Hydroxypyruvat erhalten wird, ist das chemisch dargestellte (α,β-Dihydroxyethyl)-Thiamindiphosphat nicht in der Lage, als Donorsubstrat für das Akzeptorsubstrat Erythrose-4-phosphat zu dienen. Die Strukturen von verschiedenen Transketolase-Ligand-Komplexen wurde mit Hilfe von Röntgenkleinwinkelstreuung aus Synchrotronstrahlung untersucht. Kinetische und spektroskopische Daten zeigten, dass die Bindung des Donorsubstrates Hydroxypyruvat an Transketolase zu einer Akkumulation des a-Carbanions/ Enamins, dem zentralen Intermediat der enzymatischen Thiaminkatalyse, führt. Die dreidimensionale Struktur dieses Intermediates wurde mit Hilfe von Kryokristallographie mit einer Auflösung von 1.9 Å erhalten. Die erhaltene Elektronendichte am C2-Atom des Kofaktors weist auf eine planare Struktur hin, die das Enamin des Intermediates beweist. Das Reaktionsintermediat wird durch direkte Wasserstoffbrückenbindungen zu den Histidinen 103 und 481 und über eine indirekte Wasserstoffbrücke, über ein Wassermolekül, zum Histidin 69 fixiert. Die 4'-NH2-Gruppe des Aminopyrimidinringes des ThDP befindet sich in einem Abstand von 3 Å zur α-Hydroxylgruppe des Glycolrestes, aber der Winkel ist für eine starke Wasserstoffbrücke ungeeignet. Es konnten keine strukturellen Änderungen des Transketolasemoleküls im intermediären Zustand gegenüber der Holoform des Enzyms detektiert werden, was Konformationsänderungen der aktiven Zentren während der Katalyse unwahrscheinlich macht. Das Intermediat ist mit hoher Okkupanz in beiden aktiven Zentren nachweisbar.
The cleavage of the donor substrate D-xylulose 5-phosphate by recombinant wild type and the variant H263A of yeast transketolase was studied using enzyme kinetics and circular dichroism spectroscopy. The enzyme species are able to catalyze the cleavage of donor substrates, the first half of the reaction cycle, even in the absence of any acceptor substrate. However, an increase in the rate constant for the release of the cofactor-bound glycolyl moiety ("active glycolaldehyde" or α-carbanion/enamine intermediate) was observed for the variant H263A and related to a stabilization of this intermediate by histidine 263. Chemically synthesized intermediate D, L-(α,β-dihydroxyethyl) thiamin diphosphate) is bound to wildtype transketolase with an apparent KD comparable to that of the native cofactor and calculated from titration experiments using circular dichroism spectroscopy. Both enantiomers are cleaved by the enzyme at different rates. In contrast to the enzyme-generated α-carbanion of (α,β-dihydroxyethyl) thiamin diphosphate formed by decarboxylation of hydroxypyruvate, the synthesized D, L-(α,β-dihydroxyethyl) thiamin diphosphate did not work as donor substrate when erythrose 4-phosphate is used as acceptor substrate. The structures of different transketolase-ligand complexes were investigated using small-angle x-ray solution scattering. Kinetic and spectroscopic data indicated that addition of the donor substrate hydroxypyruvate to the thiamin diphosphate dependent enzyme transketolase led to the accumulation of the a-carbanion/enamine of (α,β-dihydroxyethyl) thiamin diphosphate, the key reaction intermediate in enzymatic thiamin catalysis. The three-dimensional structure of this intermediate trapped in the active site of yeast transketolase was determined to 1.9 Å resolution using cryocrystallography. The electron density suggests a planar a-carbanion/enamine intermediate having the E-configuration. The reaction intermediate is firmly held in place through direct hydrogen bonds to His103 and His481, and an indirect hydrogen bond via a water molecule to His69. The 4'-NH2 group of the amino-pyrimidine ring of ThDP is within 3 Å distance to the α-hydroxy oxygen atom of the glycol moiety, but at an angle unfavorable for a strong hydrogen bond. No structural changes occur in transketolase upon formation of the reaction intermediate suggesting that the active site is poised for catalysis and conformational changes during the enzyme reaction are not very likely. The intermediate is present with high occupancy in both active sites.
