Zusammenfassung: Dual specificity tyrosine phosphorylation regulated kinase 3 (DYRK 3) zählt zur Kinase-Familie der DYRK-Proteine, die eine Rolle bei Wachstum und Entwicklung spielen. DYRK 3 fungiert hier als für die erythroide Linie spezifischer Inhibitor der Entwicklung von roten Blutzellen. Diese Funktion wurde sowohl für das humane als auch das Maus-Protein gezeigt. DYRK 3 enthält eine zentrale Kinase-Domäne gemeinsam mit 20 Aminosäuren, die einzig bei Kinasen der DYRK-Familie konserviert sind (DH-Box). Zudem enthält das Protein einen kurzen C-Terminus sowie eine ausgedehnte N-terminale Domäne mit bisher unbekannter Funktion. (Diese enthält 184 bzw. 164 Aminosäuren in den Isoformen DYRK 3-L und S.) Bisher ist die Funktion von DYRK 3 nur wenig verstanden. Um DYRK 3 innerhalb der Zelle zu lokalisieren, wurden verschiedene Säugetier-Zellen mit Fusionen aus DYRK 3 und GFP oder FLAG-Tag transfiziert. Durch Fluoreszenz-Mikroskopie und Zell-Fraktionierung konnte gezeigt werden, dass sich DYRK 3-S sowohl im Cytoplasma als auch im Kern von HEK-293-Zellen befindet. GFP-DYRK 3-S kann mit 91,4 kDa nicht mehr durch einfache Diffusion in den Kern gelangen. Das Protein muss also in der Lage sein, aktiv zwischen Cytoplasma und Zellkern hin und her zu wandern (Shuttling). Im Gegensatz dazu wurde DYRK 3 mit C-terminalem GFP oder N-terminalem FLAG-Epitop in HEK-293-Zellen vom Kern ausgeschlossen. Nach Deletion des N-Terminus bis zum Beginn der DH-Box befand sich auch DYRK 3-S164-568-GFP sowohl im Cytoplasma als auch im Zellkern. Der isolierte N-Terminus mit C-terminalem GFP wurde in einem Teil der Zellen vom Kern ausgeschlossen. Der Ausschluss von Zellkern war bei DYRK 3-S-GFP auch in HEP-G2-Zellen, jedoch nicht in LLC-PK1- oder COS-7-Zellen zu beobachten. Diese Ergebnisse legen nahe, dass DYRK 3 innerhalb der N-terminalen Domäne Zelltyp-spezifisch im Cytosol verankert wird. Zur Identifizierung von Wechselwirkungspartnern wurden Yeast-Two-Hybrid-Screens mit DYRK 3 als Bait durchgeführt. Einer der Klone, der mit DYRK 3 interagiert, war STAT 5A. Innerhalb des Hefe-Systems konnte der STAT 5A-bindende Teil von DYRK 3 erfolgreich eingegrenzt werden. Das N-terminale Fragment von Leu27 bis Gly184 von DYRK 3-L war für diese Interaktion ausreichend, die katalytisch aktive Kinase-Domäne von DYRK 3 war hingegen nicht für die Wechselwirkung mit STAT 5A erforderlich. Die so identifizierte Bindungsdomäne beinhaltet die gleichen C-terminalen Aminosäuren wie jenes DYRK 3-Fragment, das die Verankerung der Kinase im Cytoplasma bewirkt. Diese Bindungsdomäne von Maus-DYRK 3 wurde in E. coli als lösliches Protein exprimiert und biophysikalisch untersucht. Hierbei deuteten Analysen mittels CD-Spektroskopie und limitierter Proteolyse auf einen geringen Anteil an beta-Faltblatt und Poly-Prolin-II-Helix hin. Die Untersuchung dieses Protein-Fragments durch 15N-NMR-Spektroskopie zeigte jedoch, dass der N-Terminus von DYRK 3 zum großen Teil flexibel in Lösung vorliegt. In der vorliegenden Arbeit wurden erstmalig Hinweise für das Shuttling eines DYRK-Proteins in Säugetier-Zellen beschrieben. Darüber hinaus konnte der bisher nicht untersuchte N-Terminus von DYRK 3 für die Verankerung der Kinase im Cytosol und für die Interaktion mit dem möglichen Substrat STAT 5A verantwortlich gemacht werden. Bei der biophysikalischen Analyse dieser Bindedomäne erwies sie sich allerdings in Lösung als flexibel oder ungeordnet. Der N-Terminus von DYRK 3 ist also für die Interaktion mit anderen Proteinen von Bedeutung und beeinflusst die subzelluläre Lokalisierung von DYRK 3.
Dual specificity tyrosine phosphorylation regulated kinase 3 (DYRK 3) is a member of the DYRK family of serine/threonine protein kinases implied in growth and developmental regulation. Within the haematopoietic system, DYRK 3 plays a unique role among the DYRK family members as line specific suppressor of red blood cell development in man and mouse. DYRK 3 consists of a central kinase domain with an associated 20 N-terminal amino acids only conserved within the DYRK family of protein kinases (DH box), a short C-terminal part and a large amino acids spanning N‑terminal domain with unknown function (184 or 164 amino acids in DYRK 3-Long (L) and Short (S), respectively). However, DYRK 3 action is still poorly understood. To localize DYRK 3 within the cell, protein fusions with GFP or FLAG tag were transiently transfected into different mammalian cell lines. By fluorescence microscopy and cell fractionation studies DYRK 3-S with N-terminal GFP fusion was shown to be present within the cytosol and the nucleus. As the GFP-DYRK 3-S fusion would be restricted to the cytosol simply by its size of 91.4 kDa, the protein has to be able to shuttle between cytosol and nucleus. In Contrast, a fusion of DYRK 3 with C-terminal GFP or N-terminal FLAG tag was excluded from the nucleus of HEK 293 cells. Deletion of the N-terminal domain up to the start of the DH box caused the protein DYRK 3-S164-568-GFP to shuttle again. The N-terminal domain with C‑terminal GFP tag was excluded from the nucleus in many of the cells. Exclusion of DYRK 3-S-GFP was also clearly seen in HEP-G2 cells but not in LLC-PK1 or COS-7 cells. These findings can be explained by cell type specific cytosolic anchorage of DYRK 3 via its N-terminus. Yeast two-hybrid screens were performed to identify interacting partners of DYRK 3. Among others, one DYRK 3 interacting candidate was STAT 5A. The interaction of DYRK 3 with STAT 5A was dissected using the yeast system. Not only was the kinase activity of DYRK 3 dispensable for this interaction, but the N-terminal fragment from Leu27 to Gly184 of DYRK 3-L was sufficient for an interaction with STAT 5A. This binding domain includes the same C-terminal end as the DYRK 3 fragment causing cytoplasmic anchorage of DYRK 3-S. This binding domain of mouse DYRK 3 was then expressed in E. coli as soluble protein and characterized biophysically. Although CD and limited proteolysis studies suggested a certain content of beta sheet and poly proline II helix, the protein was shown to be flexible to a large extent by 15N NMR spectrometry. This is the first description of a mammalian DYRK protein to shuttle between nucleus and cytosol. In addition, the individual N-terminal part of DYRK 3 caused cytoplasmic anchorage of DYRK 3 and was sufficient for the interaction with STAT 5 although this binding domain was unstructured in solution. Taken together, the N-terminal part of DYRK 3 is responsible for protein-protein interactions and influences the subcellular localization of DYRK 3.
