Abstract: Es hat sich gezeigt, dass computerbasierte Methoden eine Abkürzung für die Markteinführung neuer Arzneistoffe sind. In ähnlicher Weise wurde in dieser Arbeit computergestütztes Wirkstoffdesign CADD genutzt, um die Entdeckung neuer HDAC-Modulatoren für die Behandlung verschiedener Krankheiten zu leiten. In der aktuellen Arbeit wurden mehrere computer-basierte Methoden angewandt, um neue Moleküle vorzuschlagen, die auf HDACs abzielen könnten. Erstens wurde eine neue Klasse von HDAC Inhibitoren ("Reverse-Hydroxamat" als zinkbindendes Motiv) durch virtuelles Screening entdeckt, die eine Hemmung von Schistosoma mansoni HDAC8 mit IC50-Werten im niedrigen mikromolaren bereich hemmen. Der Bindungsmodus des besten Treffers konnte durch Röntgenkristallographie bestätigt werden. Zweitens wurde eine Reihe von Molekülen mit umgekehrter HIV-Latenzzeit mittels virtuellem Screening entdeckt. Drittens wurden berechnete BFE zur Erzeugung von QSAR-Modelle benutzt, um Benzhydroxamsäuren als smHDAC8-Inhibitoren auf der Grundlage von in silico Vorhersagen weiter optimieren zu können. Gleichzeitig wurde ein Screening auf neuartige Modulatoren von HDACs aus einer Online-Datenbank für NP, die aus afrikanischen Ausgangsspezies isoliert wurden (http://african-compounds.org/anpdb/), durchgeführt. Die Analyse der ANPDB zeigte, dass sie Verbindungen enthält, die HDAC-Targets binden können.
Computational methods have been proven to be shortcuts in developing novel lead compounds and drug candidates. For example, CADD methods have been used to guide the discovery of HDAC inhibitors for the treatment of several diseases. In this thesis, several computational methods were applied to suggest novel molecules that could target HDAC isoforms. First, a new class of HDAC small inhibitor (“reverse hydroxamate” as zinc-binding motif) was discovered by virtual screening and confirmed to inhibit Schistosoma mansoni HDAC8 with IC50 values in the low micromole range. The binding mode of the best hit could be confirmed by X-ray crystallography. Second, using virtual screening, a set of molecules with reverse HIV latency was discovered. Third, a procedure of using computed BFE was developed to generate QSAR models to optimize benzhydroxamates as smHDAC8 inhibitors based on in silico predictions. Simultaneously, screening of novel modulators of HDACs from an online database for NP isolated from African source species (http://african-compounds.org/anpdb/) was carried out. Analysis of the ANPDB showed that it contain compounds that can bind to HDAC targets.
