dc.description.abstractIm Laufe des anaeroben Stoffwechsel des gram- Bakteriums Escherichia coli kann im Zuge der gemischten Säure-Gärung Pyruvat zu Formiat durch die Aktivität der Pyruvat-Formiat-Lyase (PflB) oxidiert werden. Da eine Anhäufung von Formiat im Cytoplasma zu einer Entkopplung des Protonen-Gradienten führen kann, muss ein Translokationssystem für Formiat existieren. 1994 konnte von Suppmann und Sawers das Formiat Kanal Protein FocA als potentieller Translokator identifiziert werden. Mit Hilfe von gerichteter Mutagenese wurden verschiedene Varianten von FocA überproduziert, gereinigt und mittels CD-Spektroskopie sowie Blue Native Page analysiert. Es konnten verschiedene Aminosäuren identifiziert werden, welche an der Translokation von Formiat beteiligt sind. Mit Hilfe von in vivo (bacterial two hybrid system) und in vitro (pull down Analysen, Far Western Blot Analyse) Untersuchungen konnte eine Interaktion zwischen dem Membranprotein FocA und dem cytosolischen Protein PflB nachgewiesen werden.-
dc.description.abstractDuring mixed acid fermentation in Escherichia coli pyruvate is converted to formate and acethyl-CoA by the enzyme pyruvate formate lyase (PflB). Due to the fact an accumulation of formate in the cell would lead to an uncoupling of the proton motive force, a translocation system for formate must exist. 1994 Suppmann and Sawers were able to identify the formate channel protein FocA as a potential formate translocator. By using site-directed mutagenesis different variants of FocA were overproduced, purified and analyzed by CD-spetroscopy and blue native page analysis. The influence of single aminoacids in the formate translocation could have been shown. The cytosoloic protein PflB was identified as interaction partner of the membrane protein FocA by in vivo (bacterial two hybrid system) and in vitro (pull down analysis, far western blot analysis) methods.eng
