dc.description.abstractIn den letzten Jahren hat sich das Ernährungsbewusstsein der Bevölkerung dahingehend gewandelt, dass mehr Obst und Gemüse konsumiert werden. Die darin enthaltenen bioaktiven Inhaltsstoffe wirken präventiv gegenüber Krankheiten. Ein Beispiel für diese bioaktiven Verbindungen sind Anthocyane. Auf Grund ihrer antioxidativen Eigenschaften werden sie beispielsweise zur Prävention von Krebserkrankungen des Darmes, zugeführt. Auf Grund ihrer schlechten Bioverfügbarkeit wurde im Rahmen dieser Arbeit eine Methode zur Mikroverkapselung von Anthocyanen entwickelt. Das auf Hartfett basierende mikropartikuläre System zeigte, nach der in vitro Inkubation in den Darm simulierenden Medien, eine Steigerung des Gesamtanthocyangehalts um 13 ± 2 % gegenüber nicht verkapselten Anthocyanen. Die Charakterisierung dieser entwickelten Lipidmikrokapseln mittels Lichtmikroskopie, Konfokaler Laser-Scanning-Mikroskopie, Rasterelektronenmikroskopie, Statischer Lichtstreuung und Differenzialthermoanalyse zeigte, dass sphärische Kern-Hülle Partikel mit einem wässrigen, anthocyanhaltigen Kern vorlagen. Des Weiteren wurden W1/O/W2-Doppelemulsionen in vitro mittels Pankreatinassay simulativ verdaut. Durch die Simulation des in vitro Lipidverdaus konnte gezeigt werden, dass die O-Phase der Emulsionen abgebaut wird. Die in der inneren W1-Phase verkapselten Anthocyane wurden freigesetzt. Für die Optimierung von Matrix-Mikropartikeln aus Molkenproteinisolat (MPI) wurden die rheologischen Eigenschaften von MPI bei pH-Werten von pH 1,5 und pH 6,8 bestimmt. Die ermittelten Sol / Gel-Übergangstemperaturen lagen je nach pH-Wert des untersuchten Molkenproteinisolat-Sols bei etwa 70 °C.Diese Dissertation zeigt neue Ansätze um bioaktive Inhaltsstoffe in mikropartikulären Systemen zu verkapseln und die Liberation der entwickelten Systeme zu charakterisieren. Dadurch kann der Gesamtanthocyangehalt im Darm gesteigert werden, was sich positiv auf die Absorption und die Wirkung dieser Substanzen auswirkt.-
dc.description.abstractNutritional awareness of the people has changed during the last years yielding to a higher consumption of fruit and vegetables. Bioactive ingredients of these aliments are supposed to be preventive against many diseases. One example for this bioactive compounds are anthocyanines. Due to their antioxidative properties they were used in the prevention of colon cancer. Because of their poor bioavailability, anthocyanines containing microparticles were developed in this work . These microparticulate systems were based on hard fat as wall material. Incubating these microcapsules in intestinal conditions mimicking media showed an increase of total free anthocyanin concentration of 13 ± 2 % in comparison with non encapsulated anthocyanines. Characterization of the developed microcapsules was performed by light-microscopy, confocal laser scanning microscopy, scanning electron microscopy, static light scattering and differential scanning calorimetry. With all methods spherical microparticles with a hydrophilic anthocyanin containing core were detected. Furthermore the digestion of W1/O/W2-double emulsions was simulated in vitro using a pancreatic assay. With this assay digestion of the lipid containing O-phase was demonstrated and incorporated anthocyanines of the W1-phase were released. For the optimisation of whey protein based microparticles, the rheological properties of whey proteins and their sol / gel transitions at pH 1.5 and pH 6.8 were characterized. The determined sol / gel transition temperatures were at about 70 °C, dependent on pH-level. This thesis shows new approaches to encapsulate bioactive ingredients into microparticle systems and to characterize the liberation out of these particles. As a result total anthocyanin concentration in the gut was increased which has a positive effect on the absorption and health benefit qualities.eng
