Zusammenfassung: Das α-Toxin (α-Hämolysin) aus Staphylococcus aureus ist ein häufiger Infektionserreger beim Menschen. Alpha-Toxin bewirkt konzentrationsabhängig die Aggregation von Thrombozyten, die Freisetzung von Granulainhaltsstoffen aus Thrombozyten, die Aktivierung der Tyrosinkinase Src, sowie der stressinduzierten MAPKinasen p38 und JNK1/2. Src, die häufigste zelluläre Tyrosinkinase in Thrombozyten, stand dabei im Mittelpunkt unserer Untersuchungen. Nach Aktivierung der Thrombozyten durch α-Toxin wird Src durch die Kalzium-abhängige Protease Calpain degradiert und kann demnach durch Reduktion der Konzentration von extrazellulärem Kalzium oder durch den Calpaininhibitor Calpeptin gehemmt werden. Inhibitoren der Familie der Src-Kinasen wie PP1, PP2, SU6656 und Dasatinib hemmen die α-Toxin-induzierte Aktivierung der stressinduzierten MAPKinasen p38 und JNK1/2 und die Tyrosinphosphorylierung. Der Tyrosinphosphorylierung einer Reihe von Proteinen folgt konzentrationsabhängig eine rasche, nahezu vollständige Dephosphorylierung.
A major virulence factor in the pathogenesis of Staphylococcus aureus infection is α-toxin (also denoted as α-hemolysin). Staphylococcal α-toxin induces aggregation of platelets in a concentration-dependent manner, secretion of platelet contents, activation of tyrosine kinase Src as well as activation of the two stress-activated MAPkinases p38 and JNK1/2. Regarding individual protein kinases, the most abundant cellular tyrosine kinase Src becomes proteolytically cleaved to degradation products typical for the calcium-dependent protease calpain. The cleavage of Src can be blocked with the calpactin inhibitor calpeptin or by depletion of extracellular calcium. Pre-incubation of platelets with inhibitors of the Src tyrosine kinase family PP1, PP2, SU6654 and dasatinib leads to reduced activation of the two stress-activated MAPkinases p38 and JNK1/2, as well as protein tyrosine phosphorylation. The maxima of tyrosine phosphorylation and the onset of dephosphorylation of most of the affected proteins are concentration-dependent.
