Zusammenfassung: Die folgende Arbeit befasst sich mit der Fragestellung, ob das von Schwarzkopf et al. generierte GNE+/- Mausmodell ein Modell für die Einschlusskörpermyopathie (GNE Myoptahie) darstellen kann. Es wurden insgesamt 38 Mäuse untersucht, davon 19 GNE+/- Mäuse und 19 Kontrolltiere. Die Tiere wurden zusätzlich in je zwei Gruppen unterteilt, junge, 24 Wochen alte Mäuse und alte, 80 Wochen alte Mäuse. Die Muskeln wurdensowohl histologisch als auch elektronenmikroskopisch nach GNE Myopathie typischen Merkmalen untersucht. Zudem wurden Phänotyp, Gesamtgewicht der Tiere als auch Einzelgewichte von M. tibialisanterior, M. gastrocnemius und M. quadricepsfemoris erfasst, die Kreatinkinase und Sialinsäurelevel bestimmt als auchdie Leistungsfähigkeit der Tiere mittels Laufradtest ermittelt. Phänotypisch, histologisch und elektronenmikroskopisch waren keine signifikanten, GNE Myopathietypischen Merkmale nachweisbar, es fanden sich jedoch reduzierte Sialinsäurelevel bei den jungen GNE+/- Tieren, die sich im Alter anglichen. Zudem wiesen GNE+/- Mäuse eine signifikante, um 22% verminderte Leistungsfähigkeit bis Tag vier im Laufradtest auf. Das GNE+/- Mausmodell stellt aufgrund von fehlenden histologisch und elektronenmikroskopisch nachweisbaren GNE Myopathie typischen Strukturen kein Modell für die GNE Myopathiedar. Ein neuer Aspekt ist jedoch die zunehmende Sialisierung im Alter, dessen Funktionalität bislang noch unklar ist.
This thesis deals with the question whether the C57BL/6 GNE+/- mouse modelwhich was generated from Schwarzkopf et al., could be a model of the inclusion body myopathy (GNE myopathy). Therefore 38 mice were examined, 19 GNE+/- mice and 19 control mice that did not have the mutation. They were then put into two groups each, 24 weeks and 80 weeks old mice. The muscles were analysed regarding GNE myopathy typical changes in histology and electron microscopy. Also the total weight and the weight of the single muscles, M. tibialis anterior, M. gastrocnemius and M. quadriceps femoris were examined. Serum creatine kinase and levels of sialic acids and the treadmill performance were tested. Phenotypically, histologically and in electron microscopy there were no significant GNE myopathy typical characteristics found, though there were reduced levels of sialic acids in young mice which increased with advancing age. In addition there was a significantly reduced performance in treadmill exercise within the GNE+/- mice group until day four. Because there were no GNE myopathy typical structures found, neither histologically nor in electron microscopy, the GNE+/- mouse model cannot be considered as a model for the GNE myopathy. The increasing levels of sialic acids with advancing age represent a new aspect of which the functionality is still unknown.
