dc.description.abstractDas Nierenzellkarzinom (NZK) ist die 10. häufigste Tumorerkrankung in den Westlichen Industriestaaten. Hauptursache für die Entstehung des NZK ist der Verlust des Von-Hippel-Lindau Tumorsuppressor Proteins. Dieser Verlust resultiert in der Überproduktion von Wachstumsfaktoren. Zielgerichtete Therapieansätze, wie die Gabe von Tyrosinkinaseinhibitoren (TKI), sollen die Wirkung der Wachstumsfaktoren unterdrücken. Was die Inhibierung in den Tumorzellen bewirkt und wie sie auf Tumorinfiltrierende Lymphozyten einwirken, ist nicht hinreichend erforscht. In dieser Arbeit wurden unter TKI-Behandlung anti-apoptotische Proteine herunter reguliert gefunden. Untersuchungen des Proteoms, wiesen zudem auf eine Regulation der Proteinfaltungsmaschinerie hin. Zudem wurde untersucht, welchen Einfluss die TKI auf humane Lymphozyten, speziell T-Zellen haben. Untersuchungen an PBMCs zeigten einen negativen Einfluss von Sorafenib und Sunitinib zytotoxischen T-Zellen. Axitinib-Behandlung führt zu einem G2/M-Arrest. Untersuchungen zur Resistenzentwicklung weisen auf eine Regulation der Endothelialen-Mesenchymalen-Transition (EMT) hin.-
dc.description.abstract1 of 10 tumors diagnosed in the western world are renal cell carcinoma (RCC). The main cause of RCC is the loss of the Von-Hippel-Lindau tumor suppressor protein. This causes an overproduction of growth factors. Targeted therapies, like tyrosine kinase inhibitors, inhibit corresponding growth factor receptors. What the inhibition causes in the tumor cells and what influence they have on tumor infiltrating lymphocytes is not well understood. In this work anti apoptotic proteins have been found down-regulated in TKI treated cells. Analysis of the proteome hinted to an influence on the protein folding machinery. The influence of TKI on lymphocytes, especially T cells, has been analyzed as well. Analysis of PBMCs showed a negative influence of Sorafenib on cytotoxic T cells. Axitinib treatment results in a G2/M arrest in T cells. Analysis of resistance development showed a possible regulation of endothelial-mesenchymale-transition (EMT).eng
