Abstract: In dieser Arbeit wurde die transkriptionelle Regulation des Kohlenstoffmetabolismus in der Milchhefe Kluyveromyces lactis untersucht. Ein Schwerpunkt wurde auf die primäre transkriptionelle Antwort der Zelle bei Glukosemangel gelegt. Dabei wurde die Verwertung von Ethanol und Glycerol verglichen, die teilweise sehr deutliche Unterschiede (z.B. Induktion von Genen des Methylcitratzyklus) zeigt. Weiterhin wurde der Einfluss des Snf1-abhängigen Transkriptionsfaktors (TF) Adr1 analysiert. Hierbei zeigte sich, dass sich die Bedeutung des TFs für die kohlenstoffregulierte Transkription in K. lactis größtenteils divergent von der in S. cerevisiae entwickelt hat. KlAdr1 ist zudem als negativer Regulator an der Glycerolrepression von Genen des Glyoxylatzyklus beteiligt. Im dritten Teil der Arbeit wurde die transkriptionelle Regulation des Gluconeogenese-Schlüsselenzyms KlFbp1 untersucht. Durch Promotordeletionsanalysen konnte ein cis-regulatorisches Element im KlFBP1-Promotor identifiziert (UASKlFBP1) und KlReb1 als daran bindendes Protein isoliert werden. Diese Bindung konnte durch in vitro-Analysen bestätigt werden.
In this work transcriptional regulation of carbon metabolism in the milk yeast Kluyveromyces lactis was investigated. One focus was placed on the primary transcriptional response of the cell while glucose depletion. The utilization of ethanol and glycerol, which partly shows very marked differences (e.g., induction of methyl citrate cycle genes), was compared. Furthermore, the influence of the Snf1-dependent transcription factor (TF) Adr1 was analyzed. It was shown that the importance of the TF for the carbon-regulated transcription in K. lactis has largely diverged from that in S. cerevisiae. KlAdr1 is also involved as a negative regulator in the glycerol repression of genes of the glyoxylate cycle. In the third part of this study the transcriptional regulation of the gluconeogenic key enzyme KlFbp1 was investigated. A cis-regulatory element of the KlFBP1 promoter (UASKlFBP1) was identified by promoter deletion analysis and KlReb1 was isolated as a UAS-binding protein. This binding was confirmed by in vitro-assays.
