Zusammenfassung: Gegenstand unserer Untersuchungen war die funktionelle Charakterisierung mitochondrialer Chaperone. Zu Beginn der Arbeit war lediglich ein mitochondriales Hsp70, Ssc1p, bekannt. Nach Identifizierung mitochondrialer Homologe zu bakteriellem DnaJ und GrpE, Mdj1p und Mge1p, wurde die Rolle dieser Co-Chaperone von Ssc1p in Mitochondrien untersucht. Durch genetische und zellbiologische Untersuchungen konnten wir zeigen, daß Mdj1p i) die Faltung neu in die mitochondriale Matrix importierter Proteine stimuliert, ii) eine Rolle bei der Biogenese von Proteinen spielt, die vom mitochondrialen Genom kodiert werden, iii) eine protektive Funktion bei Hitzestress ausübt, da thermolabile Protein durch Mdj1p transient vor Aggregation geschützt werden und iv) an der Replikation der mitochondrialen DNA beteiligt ist. Bei diesen Prozessen wirkt Mdj1p anscheinend nicht sehr spezifisch, da mehrere nicht-mitochondriale DnaJ-Proteine die Funktionen von Mdj1p komplementieren können. Für das mitochondriale GrpE-Homolog, Mge1p, konnten wir zeigen, daß es i) beim mitochondrialen Protein-Import benötigt wird, indem es die Wechselwirkung von Ssc1p mit in die Matrix translozierenden Polypeptidketten kontrolliert und ii) an Faltungsreaktionen in der mitochondrialen Matrix beteiligt ist. Für Ssc1p konnten wir bestätigen, daß dieses Hitzeschockprotein die Sortierung von Cytochrom b2 in den Intermembranraum kontrolliert.
Focus of the work was the functional characterization of mitochondrial chaperones. At the beginning of our studies only a mitochondrial Hsp70 protein, Ssc1p, was known. After identification of mitochondrial DnaJ and GrpE homologues, Mdj1p and Mge1p, respectively, the role of these co-chaperones of Ssc1p was elucidated. Via genetic and cell biological techniques we could show that Mdj1p i) stimulates folding of newly imported proteins in the mitochondrial matrix, ii) plays a role during biogenesis of proteins encoded by the mitochondrial genome, iii) has a protective role during heat stress as thermolabile proteins are transiently prevented from aggregation and iv) is involved in mitochondrial DNA replication. The role of Mdj1p appears not to be very specific as several non-mitochondrial DnaJ proteins can complement Mdj1p. For Mge1p we could demonstrate that i) it is required for mitochondrial protein import by modulating the capability of Ssc1p to interact with translocating polypeptides and ii) that it is involved in folding reactions in the mitochondrial matrix. For Ssc1p we could confirm that the protein controls sorting of cytochrome b2 to the intermembrane space.
