In chloroplasts, the Tat translocation system mediates the transport of fully folded proteins of various shapes across the thylakoid membrane without collapse of the membrane potential. To be able to study the mechanism of translocation, Tat translocase should be reconstituted into artificial membrane systems. In this work it was investigated whether so-called receptor complexes, which are composed of the membrane proteins TatB and TatC, can be used as a starting point for this purpose. The solubilization, characterization, recovery and reconstitution of TatBC receptor complexes from thylakoids were analyzed. Solubilization leads to a gradual dissociation of TatB from the complexes, ultimately leaving a pure TatC oligomer. Bound proteins and calcium ions inhibit this process. Subsequently, a procedure for enrichment of the complexes was developed and finally reconstitution of TatC oligomers into artificial membranes was demonstrated - a crucial step for future reconstitution of Tat translocase.
