Abstract: Das pflanzenpathogene Bakterium Xanthomonas campestris pv. vesicatoria transloziert Effektorproteine mit Hilfe eines Typ III-Sekretionssystems (T3S-Systems) in pflanzliche Wirtszellen. In der vorliegenden Arbeit wurden Kontrollproteine und konservierte Komponenten des T3S-Systems von X. campestris pv. vesicatoria funktionell charakterisiert. Die Ergebnisse zeigen, dass das Kontrollprotein HpaC einen Wechsel der T3S-Substratspezifität induziert, welcher vermutlich mit Hilfe der C-terminalen cytoplasmatischen Domäne des konservierten HrcU-Proteins erfolgt. Die effiziente Sekretion von Effektorproteinen ist von dem generellen T3S-Chaperon HpaB abhängig, das mit Effektorproteinen und der cytoplasmatischen ATPase HrcN interagiert, welche HpaB-Effektorproteinkomplexe dissoziiert. Die Chaperonaktivität von HpaB wird vermutlich durch den translozierten Regulator HpaA kontrolliert, welcher an HpaB bindet und damit die effiziente Sekretion von extrazellulären Komponenten des Sekretionsapparates ermöglicht.
The plant pathogenic bacterium Xanthomonas campestris pv. vesicatoria utilizes a type III secretion (T3S) system to translocate effector proteins into plant cells. In the present study, control proteins and conserved components of the T3S system from X. campestris pv. vesicatoria were characterized. The results show that the control protein HpaC induces a switch in the T3S substrate specificity that is mediated by the C-terminal cytoplasmic domain of the conserved HrcU protein. The efficient secretion of effector proteins depends on the general T3S chaperone HpaB, which interacts with effectors and the cytoplasmic ATPase HrcN that dissociates HpaB-effector complexes. The chaperone activity of HpaB is presumably controlled by the translocated regulator HpaA, which binds to HpaB and thus promotes the efficient secretion of extracellular components of the T3S system.
