Abstract: Die Vorhersage von kleinen Introns im Drosophila Genom führte zur Identifikation von sieben neuen Transkripten. Fünf Transkripte erweiterten bereits annotierte Gene und mlncRNA66A2 und mlncRNA42E5-1 wurden als neue putativ nicht-kodierende mlncRNAs identifiziert. Die erzeugte mlncRNA42E5-1-Nullmutante zeigte einen rezessiven Suppressoreffekt auf white-Gensilencing bei In(1)wm4h. Außerdem konnten potenzielle Zielgene des Gens, z.B. Tsp42Ee und Tpl94D, identifiziert werden. Des Weiteren scheint Hormonsignalen eine entscheidende Bedeutung bei der Differenzierung des Chromatins zuzukommen, da der Ecdysonrezeptorkoaktivator TAIMAN als Su(var) identifiziert wurde. Eine verringerte Expression der H3K4me3-spezifischen Demethylase LID scheint ursächlich für veränderte Chromatinmodifizierungen zu sein. Analysen mit einem EcRE-lacZ Transgen zeigten erstmals, dass Ecdyson-Signalling schon sehr früh in der embryonalen Entwicklung wirkt und vom Kofaktor TAIMAN abhängig ist. Es wurden eine Vielzahl an TAIMAN kontrollierten Genen identifiziert, darunter viele Gene mit Funktionen der Transkriptionsregulation und Dosiskompensation.
The prediction of small introns in Drosophila leads to the identification of 7 new transcripts. Five elongated already annotated genes, but mlncRNA66A2 and mlncRNA42E5-1 got identified as new putative non-coding mlncRNAs. The mlncRNA42E5-1 nullmutation is a recessive suppressor of In(1)wm4h system, which indicates a role in heterochromatin establishment. Potential target genes like Tsp42Ee and Tpl94D could be identified with RNASeq. In addition hormone signals seam to have a central role in establishing differentiated chromatin, because the ecdysone receptor coactivator taiman is a dominant Su(var) gene. Changed embryonic histonemarks might be due to reduced expression of H3K4me3 specific demethylase LID. Analysis with a transgene carrying ECRE-controlled lacZ gene demonstrated for the first time, that ecdysone-signalling is active very early in embryonic development and is depending on the cofactor TAIMAN. Via microarray analysis many TAIMAN controlled genes could be identified. They included a lot of transcriptional regulators, factors of the dose compensation and axis formation.
