Abstract: Die humane Parathormon-Rezeptorfamilie besteht aus den Peptidliganden Parathormon (PTH), Parathormon-related Protein (PTHrP) und Tuberoinfundibular Peptid aus 39 Aminosäuren (TIP 39), sowie den Klasse B G-Protein gekoppelten Rezeptoren (GPCRs) Parathormon-Rezeptor 1 und 2 (PTHR1, PTHR2). Für eine strukturelle und funktionelle Charakterisierung der einzelnen Moleküle, konnte hier mit Hilfe der SUMO-Fusionsmethode, erstmalig eine leistungsstarke rekombinante Herstellung von PTH (1-84), PTH (15-34), TIP 39 und der N-terminalen extrazellulären Domäne des PTHR2 (ECD-PTHR2) erreicht und diese für Ligandenbindungsstudien, sowie NMR-spektroskopische Analysen zugänglich gemacht werden. Aufbauend auf dieser Grundlage wurde ein neuer Weg zur GPCR-Regulation, durch posttranslationale Phosphorylierung der Liganden identifiziert und neue Sturktur- und Funktionsbeziehungen für ECD-PTHR2 gegenüber ihren spezifischen Liganden PTH und TIP 39 charakterisiert. Damit ist eine Voraussetzung für eine zukünftige detailierte Analyse von Rezeptor-Ligand-Wechselwirkungen und Selektionsprozessen für GPCRs geschaffen.
The three peptide ligands parathyroid hormone (PTH), the related protein (PTHrP) and tuberoinfundibular peptide (TIP 39) and the two class B G-protein coupled receptors (GPCRs) parathyroid hormone receptor 1 and 2 (PTHR1, PTHR2) compose the human parathyroid hormone receptor family. As foundation to characterise the interaction of PTHR1 and PTHR2 as well as their ligands on a molecular and structural height, within this thesis protocols were developed which allow a recombinant and high performance production of PTH ligands, TIP 39 and of the N-terminal extracellular domain of PTHR2 (ECD-PTHR2) by using SUMO-fusion approach. This important prerequisite made molecules suitable for ligand binding studies and NMR spectroscopic characterisation. In order a novel possebility for GPCR regulation was identified, via posttranslationally modification of ligands. Additionally the new structural and functional insights of ECD-PTHR2 bind to its ligands PTH and TIP 39 will help to receive detailed comprehension for receptor binding mechanism and selectivity at GPCR signaling.
