Abstract: Die Regulation des Transkriptionsfaktors Gal4 in der Milch- und Bäckerhefe weist viele Gemeinsamkeiten auf, unterscheidet sich jedoch hinsichtlich der subzellulären Lokalisation des Repressors Gal80 sowie dessen Interaktion mit den signaltransduzierenden ProteinenScGal3 (S. cerevisiae) bzw. KlGal1 (K. lactis). In dieser Arbeit wurde durch Proteinverkürzungen, Mutationen und Fusionsproteine eine N-terminale Kernlokalisationssequenz (NLS) in KlGal80 identifiziert. Zwei Aminosäureaustausche innerhalb dieser NLS führten zusätzlich zu einer gestörten Interaktion mit KlGal1. Dies zeigt zum ersten Mal die Beteiligung des KlGal80-N-Terminus an der Interaktion mit KlGal1 und die Überlappung dieser Region mit der NLS. Basierend auf strukturellen Daten war diese Interaktion unerwartet und spielt, wie hier gezeigt, für die ScGal80-ScGal3-Interaktion keine Rolle. Es wird ein Modell vorgeschlagen, nachdem der N-Terminus der Inhibierung der Kinasefunktion von KlGal1, welche in ScGal3 fehlt, dient.
The regulation of the transcription factor Gal4 in milk and bakers yeast exhibits many similarities but differs regarding the subcellular localization of the repressor Gal80 and its interaction with the signal transducing proteins ScGal3 (S. cerevisiae) and KlGal1 (K. lactis), respectively. During this work generating fragmented, mutated and fusion proteins of KlGal80 led to the identification of its nuclear localization sequence (NLS). Two amino acid exchanges in this NLS additionally led to a disturbed interaction with KlGal1. This shows for the first time the contribution of the KlGal80-N-terminus to the KlGal1-interaction and an overlap of this region with the NLS. Based on structural data this interaction was unexpected and plays, as shown here, no role in the ScGal80-ScGal3-interaction. A model is proposed where the N-terminus is necessary for the inhibition of KlGal1’s kinase activity, which ScGal3 lacks.
