dc.description.abstractDas passive Diffusionsporin PorB aus Neisseria meningitidis ermöglicht den essentiellen Membrantransport von löslichen Metaboliten. Als PAMP hat es Einfluss in der Auslösung der humoralen Immunantwort, einer Toll-like Rezeptor 2 abhängigen Reaktion. Zusätzlich verleiht es dem Bakterium pathogene Eigenschaften und ist am bakteriellen Antibiotikaresistenzmechanismus beteiligt. PorB-Kristallstrukturen konnten in Kombination mit Molekulardynamik-Analysen spezifische Kationenbindestellen, sowie das Vorhandensein von separierten Ionentransportwegen im Kanal identifizieren. Durch die gemeinsame Strukturaufklärung einer PorB Serotyp B loop 7-Variante sowie des Wildtyps, konnten Hinweise auf den PorB-TLR2-Interaktionsmechanismus identifiziert werden. Durch PorB loop 3-Varianten wurde der Einfluss spezifischer Aminosäurereste auf den Antibiotikatranslokation untersucht.-
dc.description.abstractThe passive diffusion porin PorB from Neisseria meningitides facilitates the essential membrane transport of soluble metabolites. As a PAMP, it has influence on the activation of the humoral immunity, a Toll-like receptor 2 dependent reaction. In addition, it has pathogenic features and is involved in the bacterial antibiotic resistance. PorB crystal structures together with molecular dynamics simulations identified specific cation binding sites and the presence of separate ion transport pathways in the channel. The structures of a PorB serotype B loop 7 mutant and the wildtype gave evidence on the PorB-TLR2 interaction mechanism.
